ABSTRACT

This study combined the herbal pair Platycodonis Radix-Curcumae Rhizoma(PR-CR) possessing an inhibitory effect on tumor cell proliferation and metastasis with the active component of traditional Chinese medicine(TCM) silibinin-loaded nanoparticles(NPs) with a regulatory effect on tumor microenvironment based on the joint effect on tumor cells and tumor microenvironment to inhi-bit cell metastasis. The effects of PR-CR on the cellular uptake of NPs and in vitro inhibition against breast cancer proliferation and metastasis were investigated to provide an experimental basis for improving nanoparticle absorption and enhancing therapeutic effects. Silibinin-loaded lipid-polymer nanoparticles(LPNs) were prepared by the nanoprecipitation method and characterized by transmission electron microscopy. The NPs were spherical or quasi-spherical in shape with obvious core-shell structure. The mean particle size was 107.4 nm, Zeta potential was-27.53 mV. The cellular uptake assay was performed by in vitro Caco-2/E12 coculture cell model and confocal laser scanning microscopy(CLSM), and the results indicated that PR-CR could promote the uptake of NPs. Further, in situ intestinal absorption assay by the CLSM vertical scanning approach showed that PR-CR could promote the absorption of NPs in the enterocytes of mice. The inhibitory effect of NPs on the proliferation and migration of 4T1 cells was analyzed using 4T1 breast cancer cells and co-cultured 4T1/WML2 cells, respectively. The results of the CCK8 assay showed that PR-CR-containing NPs could enhance the inhibition against the proliferation of 4T1 breast cancer cells. The wound healing assay indicated that PR-CR-containing NPs enhanced the inhibition against the migration of 4T1 breast cancer cells. This study enriches the research on oral absorption of TCM NPs and also provides a new idea for utilizing the advantages of TCM to inhibit breast cancer metastasis.

ABSTRACT

@#Objective To construct luciferase reporter plasmids of truncated fragments of different lengths of human guanylate binding protein 5(GBP5)gene promoter and analyze the transcriptional activity of each fragment to determine the core regulatory region.Methods GBP5promoter sequence was amplified by PCR,truncated into five fragments of different lengths and connected to pGL3-basic plasmid.The constructed recombinant plasmids pGL3-GBP5-11/21/31/41/51were transfected into 293FT cells and detected for luciferase activity.The binding sites of transcription factors in GBP5promoter region were predicted by JASPAR software,and Yin-Yang transcription factor 1(YY1)targeting the core regulatory region was selected and verified for the transcriptional regulatory activity.The CDS sequence of YY1 was amplified by PCR to construct the overexpression plasmid pIRES2-EGFP-YY1,which was then co-transfected to 293FT cells with plasmids pGL3-GBP5-21(-1 623 ~ +47 bp)and internal reference plasmid pRL-CMV,and detected for luciferase activity to analyze the regulation of transcription factor YY1 on GBP5 promoter activity.Results Colony PCR and double enzyme digestion identification proved that the plasmid of human GBP5 promoter reporter gene was correctly constructed;JASPAR software predicted that there were multiple transcription factor binding sites such as STAT1,YY1 and Foxp3 in GBP5promoter region.Double luciferase activity assay showed that pGL3-GBP5-21(-1 623 ~ +47 bp)showed the highest promoter activity,while the promoter activity of pGL3-GBP5-41(-520 ~ +47 bp)decreased significantly,suggesting that the core region of GBP5 promoter was located at upstream-1 623 ~-520 bp of 5 'UTR;Overexpression of YY1 significantly activated the GBP5 promoter activity and regulated the expression of GBP5.Conclusion The core regulatory region of human GBP5 promoter was located in upstream-1 623 ~-520 bp of the 5 'UTR,with a binding site of transcription factor YY1 existing in this region.Meanwhile,overexpression of YY1 significantly effected the activity of GBP5 promoter.

ABSTRACT Background: Objective: The study of sports biomechanics in sports medicine usually requires a special image analysis system (software) to obtain 3D kinematics data. Taking the swimming project in sports medicine as an example, 3D water images in water have always been relatively complicated and difficult. As light travels in different media, it will refract and reflect. When testing underwater movements, if only a land camera or an underwater camera is used for testing, the error caused by light refraction will be larger, which will affect the accuracy of the test data even more. Methods: Taking breaststroke movement as an example, a three-dimensional measurement method based on the Kwon3D movement analysis system is introduced. This method is different from the simple underwater camera test. It is a three-dimensional test method combining a land camera and an underwater camera. Two underwater cameras and two land cameras were used to simultaneously calibrate the water and underwater space with the same calibration frame in the experiment after analyzing and verifying the accuracy of 3D reconstruction. Results: The comprehensive reconstruction error is small, and the average relative error is less than 1%. Conclusions: The application of three-dimensional image analysis technology of vision systems in sports medicine is reasonable and worth promoting. RESUMO Antecedente: Objetivo: O estudo da biomecânica do esporte na medicina esportiva geralmente requer o uso de um sistema especial de análise de imagens (software) para a obtenção de dados cinemáticos 3D. Tomando o projeto de natação na medicina esportiva como exemplo, a análise de imagens 3D da água na água sempre foi um teste relativamente complicado e difícil. À medida que a luz viaja em diferentes meios, ela refratará e refletirá. Ao testar movimentos subaquáticos, se apenas uma câmera terrestre ou subaquática for usada para o teste, o erro causado pela refração da luz será maior, o que afetará a precisão dos dados de teste ainda mais. Métodos: Tomando o movimento de nado peito como exemplo, um método de medição tridimensional baseado no sistema de análise de movimento Kwon3D é introduzido. Este método é diferente do teste simples de câmera subaquática. É um método de teste tridimensional que combina uma câmera terrestre e uma câmera subaquática. No experimento, duas câmeras subaquáticas e duas câmeras terrestres foram usadas para calibrar simultaneamente a água e o espaço subaquático com o mesmo quadro de calibração. Depois de analisar e verificar a precisão da reconstrução 3D. Resultados: O erro de reconstrução abrangente é pequeno e o erro relativo médio é inferior a 1% Conclusões: A aplicação da tecnologia de análise de imagem tridimensional do sistema de visão na medicina esportiva é razoável e vale a pena promover. Nível de evidência II; Estudos terapêuticos- investigação dos resultados do tratamento.

Tomando como ejemplo el proyecto de natación en medicina deportiva, el análisis de imágenes de agua en 3D en el agua siempre ha sido una prueba relativamente complicada y difícil. A medida que la luz viaja en diferentes medios, se refractará y reflejará. Al probar los movimientos bajo el agua, si solo se utiliza una cámara terrestre o una cámara submarina para realizar la prueba, el error causado por la refracción de la luz será mayor, lo que afectará aún más la precisión de los datos de la prueba. Métodos: tomando como ejemplo el movimiento de la brazada, se introduce un método de medición tridimensional basado en el sistema de análisis de movimiento Kwon3D. Este método es diferente de la simple prueba de cámara subacuática. Es un método de prueba tridimensional que combina una cámara terrestre y una cámara submarina. En el experimento, se utilizaron dos cámaras submarinas y dos cámaras terrestres para calibrar simultáneamente el agua y el espacio submarino con el mismo marco de calibración. Después de analizar y verificar la precisión de la reconstrucción 3D. Resultados: el error de reconstrucción integral es pequeño y el error relativo promedio es inferior al 1%. Conclusiones: La aplicación de la tecnología de análisis de imágenes tridimensionales del sistema de visión en la medicina deportiva es razonable y vale la pena promoverla. Nivel de evidencia II; Estudios terapéuticos- investigación de los resultados del tratamiento.

ABSTRACT

ABSTRACT The comprehensive performance evaluation system of public sports service is an important part of public sports service system in China. How to objectively and comprehensively evaluate the present situation of public sports service performance in our country has always been a difficult problem in the development of public sports service. Based on the five principles of constructing the recipient satisfaction index, the PCSSI model of the comprehensive evaluation system of public sports service supply was constructed using scale design. Large-scale stadiums in seven districts of Shenyang were selected for investigation. The recipient satisfaction index model was established to compare the satisfaction index of the 7 regions, and analyze the influencing factors of the satisfaction index of public sports service supply, and put forward some suggestions for improving the service level of large stadiums and gymnasiums in Shenyang. Practice has proved that the application of the PCSSI model has a positive effect on improving the performance level of public sports service supply in China.


RESUMO O sistema abrangente de avaliação do desempenho dos serviços públicos de esportes é uma parte importante do sistema de serviço público de esportes na China. Como avaliar objetivamente e exaustivamente a situação atual do desempenho do serviço público de esportes em nosso país sempre foi um problema difícil no desenvolvimento do serviço público de esportes. Com base nos Cinco princípios de construção do índice de satisfação dos beneficiários, o modelo PCSSI do sistema de avaliação global do desempenho dos serviços públicos desportivos foi construído pelo projeto de escala. Os estádios de grande porte de sete distritos de Shenyang foram selecionados para investigação. O modelo do índice de satisfação dos beneficiários foi estabelecido para comparar o índice de satisfação das 7 regiões, e analisar os fatores que influenciam o índice de satisfação com os serviços públicos de esportes, e apresentar algumas sugestões para melhorar o nível de serviço dos grandes estádios e ginásios em Shenyang. A prática provou que a aplicação do modelo PCSSI tem efeito positivo na melhoria do nível de desempenho dos serviços públicos esportivos na China.


RESUMEN El sistema integral de evaluación del rendimiento del servicio público de deportes es una parte importante de ese sistema en China. Cómo evaluar de manera objetiva e integral la situación actual del desempeño del servicio público deportivo en nuestro país siempre ha sido un problema difícil. Sobre la base de los cinco principios para la construcción del índice de satisfacción del receptor, se construyó el modelo PCSSI del sistema de evaluación integral de la oferta de servicios deportivos públicos utilizando un diseño a escala. Se seleccionaron estadios a gran escala en siete distritos de Shenyang para su investigación. El modelo de índice de satisfacción del receptor se estableció para comparar el índice de satisfacción de las 7 regiones, analizar los factores que influyen en el índice de satisfacción de la oferta de servicios deportivos públicos y presentar algunas sugerencias para mejorar el nivel de servicio de los grandes estadios y gimnasios en Shenyang. La práctica ha demostrado que la aplicación del modelo PCSSI tiene un efecto positivo en la mejora del nivel de rendimiento de la oferta de servicios deportivos públicos en China.

Abstract INTRODUCTION: Leptin (LEP) is a peptide hormone that acts via leptin receptor (LEPR) binding. Genetic evidence from different human populations has implicated LEP/LEPR in the pathogenesis of coronary artery disease (CAD), and suggests that certain LEP/LEPR gene polymorphisms may increase the risk of CAD. The aim of this study was to assess two single nucleotide polymorphisms (SNPs) in LEP genes (rs2167270 and rs7799039) and two in LEPR genes (rs6588147, rs1137100) for association with CAD. METHODS: We enrolled 271 North Chinese Han CAD patients, and 113 healthy age- and sex-matched controls. Genomic DNA was extracted from whole blood, and the four SNPs were assessed using a MassArray system. RESULTS: The G allele frequency at rs2167270 was significantly higher among CAD cases than among controls. The AG genotype at rs7799039 was associated with a significantly decreased risk of CAD unlike the AA genotype used as the reference. The A allele was significantly associated with the CAD patient group. Interestingly, statistically significant differences in genotype and allele frequency at LEP rs2167270 and rs7799039 existed among females but not among males. CONCLUSIONS: The current study detected a significant association between genetic variations at LEP rs7799039 and rs2167270 and the risk of CAD in a north Chinese population, and revealed that LEP rs2167270 and rs7799039 gene polymorphisms might act as predisposing factors for CAD.

ABSTRACT

In the current study, schisandrin B(SchB)-loaded F127 modified lipid-polymer hybrid nanoparticles(SchB-F-LPNs) were developed to improve the inhibition of breast cancer lung metastasis. Modified nanoprecipitation method was used to prepare SchB-F-LPNs. The nanoparticles were spherical in shape with shell-core structure by TEM observation. SchB-F-LPNs showed a mean particle size of(234.60±6.11) nm with zeta potential of(-5.88±0.49) mV. XRD results indicated that SchB existed in the nanoparticles in an amorphous state. The apparent permeability coefficient through porcine mucus of F-LPNs was 1.43-fold of that of LPNs as shown in the in vitro mucus penetration study. The pharmacokinetics study showed that the C_(max) of SchB was(369.06±146.94) μg·L~(-1),(1 121.34±91.65) μg·L~(-1) and(2 951.91±360.53) μg·L~(-1) respectively in SchB suspensions group, SchB-LPNs group and SchB-F-LPNs group after oral administration in rats. With SchB suspensions as the reference formulation, the relative bioavailability of SchB-F-LPNs was 568.60%. SchB-F-LPNs inhibited the morphological change during transforming growth factor-β1(TGF-β1)-induced epithelial-mesenchymal transition. In addition, SchB-F-LPNs significantly decreased the number of metastatic pulmonary nodules in 4 T1 tumor-bearing mice, suggesting that SchB-F-LPNs may inhibit the metastasis of breast cancer. These results reveal the promising potential of SchB-F-LPNs in treatment of breast cancer lung metastasis.

ABSTRACT

<p><b>AIM</b>To explore the relationship between evoked potentials (EPs) and chronic anoxic brain damage by chronic intermittent hypoxia (CIH), and provide theory evidence for diagnosis and treatment of anoxic encephalopathy.</p><p><b>METHODS</b>BAEP and SLSEP were recorded in rat model with CIH (hypoxia group) and rat with normoxia (normal group). Morris water maze was used to observe learning and memory ability. Immunohistochemical method was used to investigate the expression levels of caspase-3 in brain tissue.</p><p><b>RESULTS</b>The peak latency (PL) of wave I, III, V and the interpeak latency (IPL) of wave III - V, I - V in BAEP in hypoxia group were much longer than that of in normal group (P < 0.05). The PL of wave N1, P1 of SEP in hypoxia group were much longer than that of in normal group (P < 0.05). In the water mase test, the escape latency (EL) of hypoxia group was much longer than normal group (P < 0.01). The number of caspase-3 positive cells in hypoxia group was much larger than that of in normal group (P < 0.05). There was a positive correlation among BAEP, SLSEP, the number of caspase-3 positive neuron and EL of water mase.</p><p><b>CONCLUSION</b>The alteration of BAEP and SLSEP has an apparent correlation with chronic anoxic brain damage. This provides theory evidence for diagnosis and treatment of anoxic encephalopathy.</p>
